Unfortunately, there is limited scientific information about the electrostatic interactions of gum tragacanth with proteins, especially the proteins that have been unfolded by physical approaches. In this regard, the main objective of this study is investigation the effect of thermal processing on the whey protein isolate (WPI) properties and their complexation with gum tragacanth. Materials and methods:The influence ofheating time (0, 5, 10, 15, 20, 25, 30 and 35 min at 80C) on the turbidity of WPI, alone and in the mixture with GT (r = 0.1, pH 4.0 and Cp 0.1 w/w) was surveyed. Then the flow behaviour and viscosity of gum tragacanth-WPI mixture solution in the shear rate of 0-100 S-1 (pH 4.0) was compared with the individual solutions of gum and protein. Since the maximum turbidity was observed in the range of 15-35°C, the impact of heating time at three levels (15, 25 and 35 min) and biopolymer mixing ratio at six levels (2:1, 1:1, 1:2, 1:5, 1:10 and 1:20) on the protein and carbohydrate compositions of precipitation and supernatant were investigated by lowry and phenol-sulphuric acid experiments, repectively. Results:Turbidity of protein solutions were significantly changed with rising the heating time such that the maximum turbidity achieved after 25 min thermal processing. While these solutions were unstable due to protein-protein interaction and formation of protein aggregates, GT addition leads to meaningful changes in the stability of protein solutions which was as a result of protein-polysaccharied interactions. Similar to protein solutions, the maximum turbidity of the complexes was achieved after 25min thermal processing. The rheological measurements showed that the mixture solution has pseudoplastic behavior with a large hysteresis loop as well as the higher viscosity rather than the blank solutions, all of which indicate the formation of electrostatic interactions between protein and polysaccharide and formation of complexes.By calculation of protein and polysaccharide in the supernatant and precipitation, the least ratio of Pr:PS was observed for the 25-min-treated WPI. Furthermore, the ratio of Pr:PS in the precipitation was increased significantly as Pr:PSratio was increased in the mixture. Conclusion:Addition ofgum tragacanth to the whey protein isolate will leads to a significant stability in the solution. Due to unfolding of WPI, using thermal processing on the whey protein isloate solution results in the meaningful increasing in the turbidity of protein solution and the efficiency of GT-WPI complex formation.}, keywords = {WPI,gum tragacanth,Turbidity,Complex,Viscosity}, title_fa = {اثر زمان اعمال تیمار حرارتی پروتئین بر ویژگی های کمپلکس صمغ کتیرا-ایزوله پروتئین آب پنیر}, abstract_fa = {سابقه و هدف: صمغ کتیرا در زمره صمغ های ترشحی با کاربردهای متنوع در صنایع غذایی و دارویی است. متأسفانه اطلاعات محدودی در خصوص اتصالات الکترواستاتیک این صمغ با پروتئین ها به خصوص پروتئین های دناتوره شده در دسترس است. به همین دلیل هدف اصلی پژوهش حاضر بررسی اثر اعمال حرارت بر ویژگی های ایزوله پروتئین آب پنیر و کمپلکس های حاصله با صمغ کتیرا می باشد. مواد و روش ها: ابتدا اثر مدت زمان اعمال تیمار حرارت دهی ایزوله پروتئین آب پنیر (0، 5، 10، 15، 20، 25، 30 و 35 دقیقه) در درجه حرارت 80 درجه سانتی گراد بر کدورت محلول های پروتئینی و کمپلکس صمغ کتیرا- ایزوله پروتئین آب پنیر (1/0 r =) در 4pH = و غلظت کلی 1/0% مورد بررسی قرار گرفت. سپس رفتار جریان و ویسکوزیته کمپلکس صمغ کتیرا- ایزوله پروتئین آب پنیر در درجه برش صفر تا 100 بر ثانیه با محلول های خالص صمغ و پروتئین مقایسه شد. با توجه به این که بیشترین میزان کدورت در محدوده حرارتی 35-15 درجه سانتی گراد مشاهده شد، اثر سه تیمار حرارتی (15، 25 و 35 دقیقه) و همچنین 6 نسبت ایزوله پروتئین آب پنیر حرارت دیده به صمغ کتیرا (2:1، 1:1، 1:2، 1:5، 1:10 و 1:20) بر میزان ترکیبات تشکیل دهنده دو فاز توسط آزمون های لوری و فنل- سولفوریک اسید مورد بررسی قرار گرفت. یافته ها: با افزایش زمان حرارت دهی، کدورت نمونه های پروتئین خالص، به طور معنی داری تغییر یافت؛ به طوری که بیشترین کدورت پس از 25 دقیقه حرارت دهی حاصل شده بود. اما این محلول ها به دلیل تشکیل توده های پروتئینی ناشی از برهم کنش پروتئین- پروتئین، بسیار ناپایدار بودند. با افزودن صمغ کتیرا به محلول پروتئین حرارت دیده به دلیل تشکیل کمپلکس های پروتئین-پلی ساکارید افزایش معنی داری در پایداری آن ها مشاهده شد (05/0 p <). به صورت مشابه در کمپلکس های پروتئین-پلی ساکارید نیز بیشترین میزان کدورت در نمونه ای که پروتئین آن به مدت 25 دقیقه حرارت داده شده بود مشاهده شد. بررسی رفتار جریان کمپلکس نشان دهنده رفتار سودوپلاستیک همراه با ایجاد حلقه هیسترسیس در منحنی های رفت و برگشت بود. همچنین افزایش چشمگیری در ویسکوزیته محلول کمپلکس نسبت به نمونه های خالص صمغ و پروتئین حاصل شد که همگی تأییدی بر ایجاد کمپلکس بین صمغ کتیرا و ایزوله پروتئین آب پنیر حرارت دیده است. محاسبات نشان داد کمترین نسبت پروتئین به صمغ در فاز رسوب مربوط به نمونه ای است که پروتئین آن به مدت 25 دقیقه در معرض حرارت 80 درجه سانتی گراد قرار گرفته است. همچنین با افزایش نسبت پروتئین آب پنیر به صمغ کتیرا در مخلوط، این نسبت در فاز رسوب نیز افزایش چشمگیری پیدا کرد. نتیجه گیری: در اثر افزودن صمغ کتیرا به محلول پروتئین پایداری زیادی در محلول های پروتئینی ایجاد می شود که حاصل برهم کنش الکترواستاتیک صمغ با پروتئین است. تیمار حرارتی ایزوله پروتئین آب پنیر باعث افزایش کدورت و در نتیجه افزایش راندمان تشکیل کمپلکس های صمغ کتیرا- پروتئین آب پنیر می شود که دلیل آن باز شدن ساختار پروتئین و در دسترس قرار گرفتن گروه های عملگرای پروتئین است.}, keywords_fa = {پروتئین آب پنیر,صمغ کتیرا,کدورت,کمپلکس,ویسکوزیته}, url = {https://fppj.gau.ac.ir/article_4841.html}, eprint = {https://fppj.gau.ac.ir/article_4841_6bf771721f77ac683f70d82e6dba0809.pdf} }
