بررسی اثر عصاره غلاف نخود فرنگی بر پایداری اکسایشی روغن آفتابگردان تحت شرایط تسریع شده

چکیده

سابقه و هدف: اکسایش لیپیدها یکی از مهم‌ترین دلایل افت کیفیت تغذیه‌ای و ویژگی‌های ارگانولپتیک مواد غذایی و همچنین ایجاد و پیشرفت بیماری‌های مختلف از طریق تشکیل رادیکال‌های آزاد می‌باشد. از این رو استفاده از ترکیبات ضد اکسایش به منظور حفاظت از مواد غذایی در برابر اکسایش ضروری است. امروزه عدم پذیرش ترکیبات سنتزی از سوی مصرف کنندگان باعث شده پژوهش‌های گسترده‌ای در زمینه کشف ترکیبات زیست فعال طبیعی و ایمن صورت گیرد. لذا در این پژوهش توانایی حفاظت کنندگی عصاره غلاف نخود فرنگی به عنوان یک عامل ضد رادیکال گیاهی در برابر اکسایش روغن آفتابگردان مورد بررسی قرار گرفته است.
مواد و روش‌ها: در ابتدا، عصاره زیست فعال غلاف نخود فرنگی توسط حلال‌های مختلف شامل آب، اتانول، استون و هگزان به دست آمد. میزان ترکیبات فنولی کل و فعالیت ضد رادیکالی عصاره‌ها به ترتیب با روش فولین- سیکالتیو و دو روش مهار رادیکال-های آزاد دی فنیل پیکریل هیدرازیل (DPPH) و هیدروژن پراکسید (H2O2) مورد ارزیابی قرار گرفتند. در ادامه، تاثیر عصاره زیست فعال غلاف نخود فرنگی حاوی بالاترین میزان ترکیبات فنولی کل و بیشترین توانایی ضد رادیکالی بر پایداری اکسایشی روغن آفتابگردان تحت شرایط تسریع شده در دمای 65 درجه سلسیوس طی 24 روز با تعیین عدد پراکسید، عدد پارا- آنیزیدین و عدد توتوکس مورد ارزیابی قرار گرفت. برای این منظور عصاره غلاف نخود فرنگی در پنج سطح (200، 400، 600، 800 و 1000 پی پی ام) و ضد اکساینده سنتزی BHT (200 پی پی ام) به روغن آفتابگردان افزوده شدند.
یافته‌ها: بیشترین میزان ترکیبات فنولی کل (04/140 میلی‌گرم معادل گالیک اسید بر گرم عصاره) و فعالیت مهار کنندگی رادیکال‌های آزاد دی فنیل پیکریل هیدرازیل (47/82 درصد) و هیدروژن پراکسید (63/10 درصد) در عصاره اتانولی غلاف نخود فرنگی به‌دست آمد. پس از گذشت 24 روز تحت شرایط تسریع شده حداکثر اعداد پراکسید، پارا-آنیزیدین و توتوکس برای نمونه کنترل به ترتیب معادل 185 میلی اکی والان اکسیژن بر کیلوگرم روغن، 37 و 407 به دست آمد. در حالیکه برای نمونه روغن حاوی عصاره اتانولی غلاف نخود فرنگی با حداقل غلظت این مقادیر معادل 2/128 میلی اکی والان اکسیژن بر کیلوگرم روغن ، 28 و 260 بود. نتایج حاکی از آن است که افزودن عصاره اتانولی غلاف نخود فرنگی با غلظت 1000 پی پی ام به روغن آفتابگردان در مقایسه با ضد اکساینده سنتزی BHTدر مقدار بیشینه مجاز (200 پی پی ام) موثرتر است.
نتیجه گیری: نتایج به دست آمده تایید کننده فعالیت ضد رادیکالی مطلوب عصاره غلاف نخود فرنگی خصوصا عصاره اتانولی می-باشد. از طرفی پایداری روغن آفتابگردان به علت مقدار بالای لینولئیک اسید مشکل به‌نظر می‌رسد اما نتایج حاکی از آن است که عصاره اتانولی غلاف نخود فرنگی به‌خوبی قادر به حفاظت از روغن آفتابگردان در برابر فرایند اکسایش می‌باشد. در نتیجه عصاره اتانولی غلاف نخود فرنگی را می‌توان به عنوان جایگزین طبیعی و موثر برای ضد اکساینده سنتزی BHT به منظور افزایش پایداری اکسایشی روغن آفتابگردان پیشنهاد نمود.

Evaluation the effect of green pea pod extract on oxidative stability of sunflower oil under accelerated conditions

چکیده [English]

Background and objectives: Lipid oxidation is one of the most important reasons for decrease in nutritional quality and organoleptic properties of foods as well as the development and progression of various diseases through the formation of free radicals. Hence, the use of antioxidant compounds to protect foods against oxidation is essential. Nowadays, extensive studies were carried out on exploration of natural and safe antioxidant compounds due to consumers concerns about safety of synthetic ones. Therefore, in this study, the protective ability of green pea pod extract as a plant antiradical agent against oxidation of sunflower oil was investigated.
Materials and methods: At first, green pea pod extract was obtained using water, ethanol, acetone and hexane. Total phenolic content (TPC) and antiradical activities of extracts were assessed using colorimetry method, and DPPH and H2O2 radical scavenging activity test, respectively. Then, effect of green pea pod extract containing the highest amount of total phenolics and maximum antiradical ability on oxidative stability of sunflower oil under accelerated conditions at 65 ºC for 24 days were evaluated through peroxide, p-anisidine and TOTOX values. For this purpose, green pea pod extract at five different concentrations (200, 400, 600, 800 and 1000 ppm) and the synthetic antioxidant of BHT (200 ppm) were added to sunflower oil.
Results: The highest TPC (140.4 mgGAE/g) and DPPH and H2O2 radicals scavenging activity (82.47 and 10.63%, respectively) was obtained for ethanolic bioactive extract from green pea pod. For control sample, the maximum values of peroxide, p-anisidine and TOTOX were 185 meq O2/kg oil, 37 and 407 after 24 days under accelerated conditions. While these values were 128.2 meq O2/kg oil, 28 and 208 for the oil containing the lowest concentration of green pea pod ethanolic extract. The results revealed that addition of green pea pod ethanolic extract at a concentration of 1000 ppm in sunflower oil was more effective than BHT (200 ppm).
Conclusion: The results obtained confirmed the suitability of green pea pod bioactive compounds antiradical activity especially for ethanolic extract. On the other hand, stability of sunflower oil seems to be difficult due to the high amount of linoleic acid but the results revealed that ethanolic extract of green pea pod was able to protect sunflower oil against oxidation as well. As a result, green pea pod ethanolic extract could be suggested as a natural and effective alternative for BHT in order to increase oxidative stability of sunflower oil.
